Genetic Studies of Paralyzed Mutants in Salmonella. II. Mapping of Three mot Loci by Linkage Analysis.

reported in the preceding paper (ENOMOTO 1966), motility genes of Aialmonella typhimurium consist of three functional units: motA, motB, and motC. motA and B adjoin each other but motC is separate and is transduced jointly with the HI gene. However, the chromosomal location of motA and B and the position of motC relative to H I have not been investigated. In order to analyse the linkage relationship between the mot loci and other genes, a system of sexual recombination is useful. In Salmonella species, none of the Hfr strains had been isolated from natural habitats. However, many investigations have been carried out by hybridization between Escherichia coli and Salmonella (BARON et al. 1959a, b; MIYAKE and DEMEREC 1959; ZINDER 1960a; MAKELA, LEDERBERG and LEDERBERG 1962; ~ K E L A 1964) and the hybrids obtained were found to recombine with Salmonella in further mating (ZINDER 1960b; DEMEREC et al. 1960; ~ K E L A et al. 1962). Furthermore, MAKELA (1 963), and SANDERSON and DEMEREC (1965) succeeded in isolating several Hfr mutants from Salmonella infected with F from E. coli K-12 and made possible overall genetic analysis in Salmonella species. In the present paper, the linkage analysis of the three mot loci in relation to other chromosomal genes by means of sexual recombination between Salmonella abony Hfr mutant and Salmonella Fis reported.
